If I understand your question correctly, DNS runs on either TCP or UDP,
on port 53, but applications and tools like nslookup and dig use UDP,
as well as the resolver libraries.

If you weren't asking about the transport, then I guess you would be
interested in DNS itself. There are a lot of RFC's that cover this. If
you don't have it already, I'd suggest getting a copy of DNS & BIND from
O'Reilly. Also, download the latest BIND source and have a look at the
docs. There have been a few recent changes (like $TTL in zone files)
that aren't discussed in the book.
